This course assumes
no prior knowledge of immunohistochemistry and introduces the participants
to concepts from the basics. It would be suitable for both the
beginner or those who would like to refresh their knowledge and practical
skills in immunohistochemistry.
It is recommended
that participants have some prior knowledge of histochemistry or have
completed the Introductory
Histotechniques course.
The course is completed
over two days and consists of a 1 hour lecture in the mornings followed
by hands on practical sessions in the laboratory for the remainder
of both days.
Concepts
covered during the lectures include:
- Fixation for
immunohistochemistry
- Direct and indirect
immunostaining
- Reporter molecules
- Endogenous enzyme
activity
- Blocking techniques
- Double labelling
- Antigen retrieval
- Choice of controls
- Trouble shooting
In the practical
sessions participants will:
- Demonstrate
smooth muscle actin using the ABC technique
- Restore vimentin
immunoreactivity using heat induced antigen retrieval
- Immunofluorescently
demonstrate smooth muscle actin using the indirect technique.
Because class size
is limited to 10, there is ample opportunity to tailor certain parts
to individual needs. Participants are encouraged to bring along
their own antibodies and tissue sections to add to their staining
runs or to bring previously attempted slides to trouble shoot with.
